Öğe A study on determination of bioactive potentials of certain lichens(Akademiai Kiado Rt., 2018) Korkmaz A.I.; Akgul H.; Sevindik M.; Selamoglu Z.Lichens are symbiotic associations that are formed by fungi and algae or cyanobacteria. The number of lichen species investigated pharmaceutically is still very low at present. The present study aims to determine the antioxidant activities, antibacterial activities, DNA protective activities, and oxidative stress status of Bryoria fuscescens (Gyeln.) Brodo & D. Hawksw., Parmelina tiliacea (Hoffm.) Hale, and Umbilicaria decussata (Vill.) Zahlbr. Lichens were extracted with ethanol in the Soxhlet device. The DPPH method was used to determine antioxidant activities. DNA protective activity was determined using pBR322 supercoil DNA. Antibacterial activity was determined with dilution test on 5 different species of bacteria (Enterocossus faecalis, Klebsiella pneumoniae, Pseudomonas aeruginosa, Escherichia coli, and Staphylococcus aureus). Total antioxidant status (TAS), total oxidant status (TOS), and oxidative stress index (OSI) were defi ned with Rel Assay Diagnostics kits. It was observed that DPPH free radical scavenging activities in lichen ethanol extracts increased with increasing concentration. The highest antioxidant activity was observed in B. fuscescens and the lowest activity was determined in U. decussata. It was also determined that the ethanol extracts of all lichen samples had DNA-protective activity. The highest antibacterial activity was detected in B. fuscescens, while the lowest activity was detected in U. decussata. It was determined that B. fuscescens had the highest oxidative stress index and U. decussata had the lowest value. It appears that the ethanol extracts of the lichen samples utilized in the study could be used as an alternative and complementary resource in medical treatment. © 2018 Akadémiai Kiadó, Budapest.Öğe Astragalin: A Bioactive Phytochemical with Potential Therapeutic Activities(Hindawi Limited, 2018) Riaz A.; Rasul A.; Hussain G.; Zahoor M.K.; Jabeen F.; Subhani Z.; Selamoglu Z.Natural products, an infinite treasure of bioactive chemical entities, persist as an inexhaustible resource for discovery of drugs. This review article intends to emphasize on one of the naturally occurring flavonoids, astragalin (kaempferol 3-glucoside), which is a bioactive constituent of various traditional medicinal plants such as Cuscuta chinensis. This multifaceted compound is well known for its diversified pharmacological applications such as anti-inflammatory, antioxidant, neuroprotective, cardioprotective, antiobesity, antiosteoporotic, anticancer, antiulcer, and antidiabetic properties. It carries out the aforementioned activities by the regulation and modulation of various molecular targets such as transcription factors (NF-B, TNF-?, and TGF-ß1), enzymes (iNOS, COX-2, PGE2, MMP-1, MMP-3, MIP-1?, COX-2, PGE-2, HK2, AChe, SOD, DRP-1, DDH, PLC?1, and GPX), kinases (JNK, MAPK, Akt, ERK, SAPK, IB?, PI3K, and PKCß2), cell adhesion proteins (E-cadherin, vimentin PAR-2, and NCam), apoptotic and antiapoptotic proteins (Beclin-1, Bcl-2, Bax, Bcl-xL, cytochrome c, LC3A/B, caspase-3, caspase-9, procaspase-3, procaspase-8, and IgE), and inflammatory cytokines (SOCS-3, SOCS-5, IL-1ß, IL-4, IL-6, IL-8, IL-13, MCP-1, CXCL-1, CXCL-2, and IFN-?). Although researchers have reported multiple pharmacological applications of astragalin in various diseased conditions, further experimental investigations are still mandatory to fully understand its mechanism of action. It is contemplated that astragalin could be subjected to structural optimization to ameliorate its chemical accessibility, to optimize its absorption profiles, and to synthesize its more effective analogues which will ultimately lead towards potent drug candidates. © 2018 Ammara Riaz et al.Öğe Determination of anti-oxidative, anti-microbial activity and heavy metal contents of Leucoagaricus leucothites(NLM (Medline), 2018) Sevindik M.; Rasul A.; Hussain G.; Anwar H.; Zahoor M.K.; Sarfraz I.; Selamoglu Z.Mushrooms, a treasure of diverse bioactive scaffolds, have been widely admired due to their nutritional and medicinal significance all over the world. The current study intended to evaluate the therapeutic potentiality of an edible mushroom, Leucoagaricus leucothites (Vittad.) Wasser. Thus, anti-oxidant potential of L. leucothites was determined using DPPH assay and for the determination of anti-microbial potential agar dilution procedure was followed. TOS (total oxidant status), TAS (total anti-oxidant status), and OSI (oxidative stress index) values were evaluated utilizing Rel Assay Kits. For the assessment of heavy metal contents, wet decomposition approach with atomic absorption spectrophotometry was adopted. Screening of phytochemicals present in ethanolic extract of L. leucothites were determined by HPLC. TAS, TOS and OSI values were found to be 8.291mmol/L, 10.797µmol/L and 0.130 respectively. Our results declared that heavy metal contents are generally in the safe range. Phytochemical analysis of L. leucothites has affirmed the presence of important phenolics such as gallic acid, catechin, and hesperidin. Investigations on anti-oxidant and anti-microbial potential of L. leucothites has uncovered the fact that this naturally occurring, biologically active, and therapeutically effective mushroom specie has natural borne anti-oxidant and anti-microbial potential and it would be worthwhile to use it for nutritional as well as medicinal purpose.Öğe Determination of some heavy metal levels in three freshwater fish in Keban Dam Lake (Turkey) for public consumption(Iranian Fisheries Research Organization, 2019) Caglar M.; Canpolat O.; Selamoglu Z.[No abstract available]Öğe Epipsammic diatoms in ağın region of keban dam lake in turkey(Green Wave Publishing of Canada, 2019) Caglar M.; Selamoglu Z.; Pala G.In this study, the epipsammic diatoms in Ağın Region of Keban Dam Lake were analyzed at two stations between March-November 2016. As a result, 41 diatom taxa in total were recorded; while two of these taxa belonged to the Centrales, 39 of them belonged to the Pennales ordo. The epipsammic diatoms showed their best growth in early months of autumn and in summer when light and temperatures were high. During the study, at both of the stations, the most significant diatoms in terms of their numbers of individuals and frequency of occurrence were Navicula spp., Nitzschia spp. and Cymbella spp. © 2019, Green Wave Publishing of Canada. The present study aims to determine and compare total antioxidant status (TAS), total oxidant status (TOS), oxidative stress index (OSI) values, and Fe, Zn, Cu, Pb, and Ni levels in Fomitopsis pinicola (Sw.) P. Karst samples gathered in Balikesir province Kazdagi National Park and Yalova province Çinarcik Hasan Baba Woods in Turkey. TAS, TOS, and OSI values of mushroom samples were measured with Rel Assay kits. Mushroom heavy metal content was determined using an atomic absorption spectrophotometer and wet decomposition procedure. In the samples collected from Çinarcik district, OSI values were 0.99±0.03, while in the samples collected from Kazdagi National Park, OSI values were 0.13±0.01. Fe content in the samples collected from Çinarcik district were 265.9±70.5 ppm, while Fe content in the samples collected from Kazdagi National Park were 31.31±1.43 ppm. As a result, it is considered that the mushrooms could be used as antioxidant source.
