    Diagnosis of the casual viruses of crop plants

    Plant viruses are major problem in agriculture and have the potential to cause significant losses. Unfortunately, there is no direct control measure or feasible and economical chemical agents that could be effective against plant viruses. The accurate detection and diagnosis of a viral disease is an important step for implementing control measures. Various diagnostic methods have been developed and are available for authentication of the diagnosis of plant viruses. The advances in the biotechnological techniques for the detection of nucleic acid and proteins have enabled to quality and reliable detection and identification of the plant viral diseases. This chapter is an overview of available methods for the effective diagnosis of plant viruses and discusses the possible advantages and limitations in their application. © 2020 Elsevier Inc.     Mulberry

    The globally important usage of mulberry trees (Morus spp.) is the production of the silkworm, and its fruits have also value as a fresh and dry fruits. Mulberry trees have affected many diseases, and the viral diseases are more prevalent in countries like China, Japan, India, Taiwan, etc. These viruses were found to be associated with mulberry mosaic disease, mulberry ringspot disease, mulberry yellow-net vein disease, and mulberry yellow streak disease. There are some reports on viral agents associated to those disease complexes and several other new occasions. The causative agent identification of viral symptoms in mulberry plants is scarcely searched and the etiological investigations remained limited. © 2024 Elsevier Inc. All rights reserved.

    Üzümsü meyvelerde hastalığa sebep olan bazı fungusların moleküler teşhisi

    Üzümsü meyve yetiştiriciliğinin önemli sorunlarından birisi de fungal hastalıklardır. Fungus hastalıklarından dolayı üzümsü meyvelerde önemli verim kayıpları yaşanmaktadır. Bu çalışmada, Doğu Karadeniz Bölgesi ve Kahramanmaraş ilindeki yabani üzümsü meyve bitkileri (çilek, ahududu, böğürtlen, maviyemiş, mürver) ve Akdeniz Bölgesi çilek yetiştiriciliği yapılan alanlardan toplanan çilek bitkileri ile PCR ve DNA dizi analizleri gibi moleküler tanı teknikleri kullanılarak fungal türler belirlenmiştir. Funguslar, PDA besi ortamında geliştirildikten sonra DNA'ları izole edilmiştir. Multi-lokus sekans tiplendirme yöntemi ile ITS ve LSU gen bölgeleri PCR ile çoğaltılmış ve elde edilen DNA dizilerinden fungal türler tespit edilmiştir. BLAST analizlerine göre, izole edilen fungusların Boeremia exigua, Colletotrichum gloeosporioides, Epicoccum nigrum, Didymella glomerata, Preussia minima, Peyronellaea prosopidis, Fusarium sambucinum, Pestalotiopsis sp, Chaetomium fimeti, Gnomoniopsis idaeicola, Hypoxylon perforatum, Diplodia seriata, Fusarium solani ve Fusarium equiseti türleri oldukları tespit edilmiştir. Bu çalışmanın bitki fungal hastalıklarla mücadele yöntemlerine katkı sağlayacağı düşünülmektedir.
