Effect of boric acid and quercetin combination on oxidative stress/cognitive function in parkinson model

Özet
Boric acid (BA) affects alveolar bone density related to mineral composition in dental physiology and nicotine (N) exposure is an important risk factor in the pathophysiology of periodontal disease. The aim of this study was to investigate the effect of BA on the deprive of bioactivity in cementoblast cells after N exposure. For this purpose, cementoblast cells were treated with different concentrations (10-1, 10-2, 10-3, 10-4, 10-5, 10-6 mM) of N and BA at a concentration of 10 ng/mL and cell viability was evaluated at 24 and 72 h by MTT assay. The migration potential of cells into the wound area was visualized by cell migration wound healing assay. Total RNA was isolated from cells on days 3, 6 and 9 and gene expression of mineralized tissue-associated markers was analyzed using real-time polymerase chain reaction. The results showed that N had a significant effect on the viability of cementoblast cells, migration-based wound healing, especially on the expression of mineralized tissue-related genes (p<0.01). In addition, it was determined that target gene expression patterns were negatively affected with the increase in N exposure on a daily basis (p<0.01), and BA application may be a powerful preventive therapeutic agent in the protection of cementoblast cell functions. © 2023, Turkish Energy Nuclear and Mining Research Institute.
