Öğe Atatürk Baraj Gölü'nde Yaşayan Carasobarbus luteus (H., 1843) ve Carassius carassius (L., 1758) Balıklarının Spermatolojik Özelliklerinin Karşılaştırılması(2014) Aral, Faruk; Doğu, Zafer; Şahinöz, ErdinçBu çalışmanın amacı, Carasobarbus luteus (Heckel, 1843) ve Carassius carassius (L., 1758) balıklarının başlıca spermatolojik özelliklerinin belirlenmesidir. Spermalar üreme sezonunda, 30 adet C. luteus ve 20 adet C. carassiustan abdominal masaj yöntemi ile alındı. Alınan spermalarda, miktar, motilite, motilite süresi, yoğunluk ve pH belirlendi. Çalışmadaki C. luteus ve C. carassius balığından alınan sperma miktarı, spermatozoa motilite oranı, spermatozoa motilite süresi, spermatozoa yoğunluğu ve sperma pHı sırasıyla ortalama 0,72±0,08 ml ve 1,03±0,20 ml; %57,67±3,67 ve %81,00±1,43; 190,30±11,25 sn ve 107,30±12,03 sn; 13,15±1,33 x109 /ml ve 10,53±1,28x109 /ml; 8,13±0,11 ve 7,60±0,04 olarak belirlendi. Söz konusu iki türün spermatozoa motilite oranı, motilite süresi, spermatozoa yoğunluğu ve sperma pHı arasında önemli farklılıklar gözlemlendi.Öğe Atatürk Baraj Gölü’ndeki (Şanlıurfa) Şabut Balığının (Barbus grypys; H, 1843) Bazı Spermatolojik ve Hematolojik Ozelliklerinin Belirlenmesi(2014) Doğu, Zafer; Aral, Faruk; Şahinöz, ErdinçBu çalışma, üreme sezonundaki Şabut balığının (B. grypus; H, 1843) bazı spermatolojik ve hematolojik özelliklerinin belirlenmesi amacıyla yapılmıştır. Araştırma, Atatürk Baraj gölü'nden yakalanan 20 adet erkek B. grypus kullanılarak gerçekleştirildi. Sperma ve kan örnekleri günlük olarak alınıp değerlendirildi. Alınan spermalarda, ortalama sperma miktarı (µL), spermatozoa motilitesi (%), motilite süresi (s), spermatozoa yoğunluğu (x109/mL) ve pH değerleri sırasıyla ortalama 886.00 ±78.76, 71.25 ±4.56, 115.10 ±6.19, 10.61 ±1.67 ve 8.13 ±0.45 olarak bulundu. Erkek B. grypus'un kan parametreleri WBC (x103/mm3), RBC (x106/mm3), Hb (g/dl), MCV (µm), MCH (pg), MCHC (%) ve PCV (%) için sırasıyla, 30.01 ±4.11, 2.05 ±0.07, 7.72 ±0.11, 147.27 ±4.93, 38.98 ±2.61, 26.47 ±0.84 ve 29.45 ±0.75 oldu. Sekiz yaşındaki balıkların sperma miktarı 5, 6 ve 7 yaşındaki örneklerden daha yüksek bulundu (P<0.05). Keza, 8 yaşındaki balıkların ortalama PCV değeri diğer yaşlardaki balıklardan daha yüksek oldu. On a effectue des examens histopathologiques et biochimiques pour mesurer les activites de la superoxyde dismutase (SOD), de la catalase (CAT), de la glutathion peroxydase (GSH-Px), et les taux de glutathion (GSH) reduit. On a divise des rats males en 3 groupes : ayant subi une operation factice (sham) (n = 12), I/R (n = 12) et I/R avec 3AB (I/R-3-AB) (n = 12). On a occlus l'artere testiculaire gauche pendant 1 h, puis on l'a soumis a une reperfusion pendant 24 h (pour les examens biochimiques et histopathologiques) et 30 jours (pour l'analyse de sperme). L'administration de 3-AB par voie intraperitoneale 10 min avant et 1 h apres la reperfusion a augmente la diminution induite par l'I/R de la motilite du sperme dans les 2 testicules, et elle a diminue l'augmentation anormale des taux de sperme dans le testicule ipsilateral. Toutefois, le traitement par 3-AB n'a pu prevenir la diminution induite par l'I/R du taux de sperme dans les 2 testicules. Les activites de SOD et de CAT sont demeurees stables chez tous les groupes. L'I/R a augmente l'activite de la GSH-Px et les taux de GSH. Le traitement par 3-AB a renverse l'augmentation induite par l'I/R de l'activite de la GSH-Px comme chez les sham, mais il n'a pas modifie les taux de GSH. Le traitement par 3-AB a augmente de maniere significative la diminution induite par l'I/R du score histopathologique. En conclusion, un traitement par 3-AB offre des avantages biochimiques et histopathologiques potentiels au-dela de la qualite du sperme et pourrait diminuer l'alteration de la torsion testiculaire.Öğe Gökkuşağı Alabahk (Oncorhynchus mykiss W., 1792)Spermasında Motilite, Canlılık ve Fertilizasyon Üzerine Bir Sulandırıcı Olarak Üzüm Pekmezinin Etkisi(2016) Temamoğulları, Füsun; Şahinöz, Erdinç; Aral, Faruk; Doğu, ZaferBu çalışmada, gökkuşağı alabalıklarının (Oncorhynchus mykiss) dondurulan spermasına 5, ve 10 oranında katılan pekmezin spermatozoa motilitesi, canlı spermatozoa oranı ve döllenme oranı üzerine etkisi incelendi. Altı adet erkek balık sperması dimetil sülfoksit ile birlikte 5, ve 10 oranında pekmez içeren sulandırıcı ile sulandırıldı. Spermalar 0.25 ml payetlerde, sıvı azot buharında dondurularak hafta boyunca sıvı azot içerisinde muhafaza edildi. Spermatozoa motilitesi üzerine pekmez etkisi açısından, %10 oranında üzüm pekmezinin (P<0.001) daha faydalı olduğu ve (P>0.05) döllenme oranı üzerine pelanezin tüm oranlarında önemli bir fark bulunamadığı saptandı. Ancak, 10 oranında kullamlan pekmez ile dondurulan sperınalardan elde edilen döllenme oranı glikoz sulandırıcısı için elde edilen en yüksek değerden daha iyi oldu (96.4%). Çalışma sonuçlarına göre, 10 oranında pekmezin gökkuşağı alabalıklarının spermalarınm dondurulmasında kullanılabileceği kanaatine varıldı.Öğe Pesticide-Contaminated Feeds in Rainbow Trout (Onchorhyncus mykiss W. 1792) Aquaculture: Oxidative Stress and DNA Damage(ZOOLOGICAL SOC PAKISTAN, 2015) Dogu, Zafer; Sahinoz, Erdinc; Aral, Faruk; Celik, Hakim; Koyuncu, Ismail; Taskin, Abdullah; Aksoy, NurtenAlthough chlorpyrifos (CPF) is an organophosphate medium-toxicity pesticide, it is highly toxic to fish and aquatic animals This study was conducted with young rainbow trouts (n=20) raised on twelve floating cages on the Ataturk Dam Lake. The fish were divided into four groups and were fed no insecticide (control groups), 0.02, 0.04 and 0.08 mg/l CFP-contaminated feed orally for 30 days. The oxidative status of trout blood tissues and the comet assay was used to measure DNA strand breaks in blood erythrocytes. No significant differences were observed at all the doses of CFP between the values of hematocrit levels and total antioxidant status. On the other hand, there was a significant correlation of increased CFP doses with leucocyte levels (F<0.05), total oxidant status (F<0.01), oxidative stress index (P<0.05) and DNA damage (F<0.01).Öğe Reproductive Characteristics of Mullet (Liza abu H., 1843) (Pisces, Mugilidae) in the Ataturk Dam Lake, Southeastern Turkey(CENTRAL FISHERIES RESEARCH INST, 2011) Sahinoz, Erdinc; Dogu, Zafer; Aral, Faruk; Sevik, Ramazan; Atar, Hasan HuseyinIn this study reproductive biology of Liza abu (Pisces Mugilidae) from Ataturk Dam Lake in the Euphrates River Basin was observed. Some reproductive characteristics such as age, egg number, sexual determination, gonadosomatic index (GSI) of Liza abu (n=821) are taken into consideration. The relationship between total weight (W(t)), and total length (L(t)) and fecundity (F) were investigated during the period from January 2004 to December 2005. The ages of investigated fish ranged from 1 to 5. The ratio of the numbers of fish samples with egg (89): total of female (413): total sample of Liza abu (810) were determined as 1:4.64:910 respectively. Though the highest gonadosomatic index (GSI) of female was found in April, male's was highest in May. The relationships between the total weight (W(t)) and fecundity (number of eggs= F), total length (L(t)) and fecundity (F) of L. abu were found as F(w) = 25.5+0.00583W(t) (R(2)=0.557) and F(L) = 14.2+0.000373 L(t) (R(2)=0.496) respectively. Relationship between total weight (W(t)) and gonad weight (W(g)) in males and females were found as W(t)=38.5+2.55W(g) (R(2)=0.222) and W(t)=36.8+4.55W(g) (R(2)=0.331), respectively.Öğe The Development of Optimal Cryopreservation Media For Longspine Scraper (Capoeta trutta) Sperm(2018) Şahinöz, Erdinç; Doğu, Zafer; Aral, FarukThis study is performed to determine some of sperm quality after applying freezing /thawing process. Thus, the aim of this study is to examine different cryprotective agentswith additives in terms of their effects at different pH on the cryopreservation process oflongspine scraper (Capoeta trutta). The present study, twelve media were prepared bymixing three different cryoprotectants (dimethyl sulfoxide (DMSO, (CH3)2SO); methanol(CH3OH); methyl glycol (MG, CH3O (CH2)2OH)) with an extenders (glucose) at fourdifferent pH (7.2, 7.6, 8.0 and 8.4) for longspine scraper semen. Considering the findingsfrom the examination (The motility rate after thawing process and duration of motilityobtained in DMSO as 81% and 20 min, in methanol as 73% and 12 min, in methyl glycolas 60% and 15 min.), we can conclude that the DMSO is the best freezing media in orderto create new essays in cryopreservation for sperm of Capoeta trutta in the future.Öğe The effect of air pressure with cervical artificial insemination on the fertility of awassi ewes synchronized with $PGF2\\alpha$(2010) Aral, Faruk; Yavuzer, Ümit; Zonturlu, Abuzer KafarBu çalışmada, İvesi koyunlarda nativ sperma ile servikal sun’i tohumlama sırasında uygulanan hava basıncının fertilite üzerine etkisi araştırıldı. Araştırmada, aşım sezonu içerisinde bulunan ve normal siklusa sahip toplam 60 baş İvesi koyun (2-5 yaşlı) kullanıldı. Tüm koyunlara bir PGF2? analoğunun kas içi iki dozunun 7 gün arayla uygulamasını içeren PG-7 gün protokolü uygulandı. Hayvanlar iki gruba ayrıldı. Birinci gruba (n=30) sevikal sun’i tohumla yapıldı. İkinci gruba (n=30) bir hava pompası aracılığıyla servikal sun’i tohumla gerçekleştirildi. Sun’i tohumlamalar ikinci PGF2? enjeksiyonundan 48 saat sonra yapıldı. Gebelikler sun’i tohumlamayı takiben 30. günde transrektal ultrasonografi ile belirlendi. Gebelik oranı, hava pompası ile yapılan servikal sun’i tohumlamada (APCAI), servikal sun’i tohumlamadan (CAI) daha yüksek bulundu (%80.0 karşılık %46.7) (P<0.01). Kuzulama oranı, hava pompası ile servikal sun’i tohumlamada, servikal sun’i tohumlama grubuna göre daha yüksek bulundu (%70.0, %46.7) ve tohumlama metodu kuzulama oranı üzerinde önemli etkiye sahip oldu (P<0.05). Koyun başına doğan kuzu oranı tohumlama metotlarından etkilenmedi (APCAI, 1.09; CAI, 1.14). Sonuç olarak, İvesi koyunlarında hava pompası ile servikal sun’i tohumlama, mekanizması tam olarak belirlenemese de klinik yönden gebelik oranını artırdığı kanaatine varıldı.Öğe The Effect of Air Pressure with Cervical Artificial Insemination on the Fertility of Awassi Ewes Synchronized with PGF2 alpha(KAFKAS UNIV, VETERINER FAKULTESI DERGISI, 2010) Aral, Faruk; Yavuzer, Umit; Zonturlu, Abuzer KafarThe aim of this study was to investigate the effect of air pressure on fertility during cervical artificial insemination with fresh semen in Awassi ewes. A total of 60 reproductively normal-cycling Awassi ewes (2-5 years old) were used in the experiment during the breeding season. All ewes were treated with the PG-7 days protocol consisting of two intramuscular doses of a PGF2 alpha analogue administered 7 days apart. Animals were divided into two groups. The first group (n=30) was cervically inseminated (CAI). The second group (n=30) was cervically inseminated by means of an air pump (APCAI). Artificial inseminations were performed 48 h after the second PGF2 alpha injection. Pregnancy was determined using ultrasonography on d 30 after AI. The pregnancy rate was found significantly higher in the APCAI group than in the CAI group (80.0% versus 46.7%) (P<0.01). The lambing rate was found to be greater in the group cervically inseminated with an air pump than in the group cervically inseminated with the conventional method (70.0 versus 46.7%). The insemination method used had a significant effect on lambing rate (P<0.05). Prolificacy was not affected by insemination methods (APCAI, 1.09; CAI, 1.14). It was concluded that cervical insemination with an air pump in Awassi ewes might improve clinical pregnancy rates by mechanisms yet to be defined.Öğe The Protective Effects of Curcumin on Organophosphate Insecticide Chlorpyrifos-Induced Oxidative Stress and DNA Damage in Oncorhynchus mykiss(Central Fisheries Research Inst, 2020) Sahinoz, Erdinc; Aral, Faruk; Dogu, Zafer; Koyuncu, Ismail; Yuksekdag, OzgurThe aim of this study was to determine the levels of enzymatic and nonenzymatic antioxidants and DNA damage in blood of rainbow trout (Oncorhynchus mykiss) exposed to chlorpyrifos (CPF) and the protective effects of curcumin (CUR) on CPF-induced oxidative stress and DNA damage. For this purpose, 0.5% and 1.0% of CUR was administered via the intraperitoneal route for 45 days during exposure to water containing 0.040 mg/L CPF in rainbow trout. We observed a significant decrease in total peroxide (TP), glutathione (GSH), GSH peroxidase (GPx), superoxide dismutase (SOD), catalase (CAT), paraoxonase (PON), and ferric ion reducing antioxidant power (FRAP) levels, whereas there was a significant increase in TP level in CPF-exposed rainbow trout (P<0.05). Antioxidants, depending on the dose, had relatively modest to significant effects on CPF-induced oxidative stress. Low doses of CUR also significantly increased serum FRAP levels in rainbow trout exposed to CPF (P<0.05). We observed increased comet assay levels in blood, predominantly in rainbow trout exposed to CPF (P<0.05). The amount of DNA damage induced by CPF exposure was more than that in the control. Overall, this study shows that CUR, depending on the dose, may modify CPF toxicity to a moderate or high degree in rainbow trout.
