Öğe Histomorphometric analysis of the effects of grape seed extract (vitis vinifera) and low-level laser therapy (LLLT) on fracture healing(2020) Ege, Bilal; Aras, Mutan Hamdi; Akgül, Hasan; Kara, Muhammed İsa; Aydemir, Işıl; İnan, Sevinç; Taysı, SeyithanAim: In this study, we aim to investigate the effects of supplementary Grape Seed Extract (GSE) and Low-Level Laser Therapy (LLLT)on fracture healing, oxidant and anti-oxidant system in experimental mandible fractures.Materials and Methods: 48 Wistar Albino rats (adult male, n=48) were used in our study. For all the subjects, a vertical fracture linethrough molar teeth in right mandibles was created and internally fixed using a four-hole microplate and four micro screws. Firstly,these subjects were randomly divided into 4 main groups (Control, GSE, LLLT, GSE+LLLLT) of 12 animals each, and then these wereindividually separated into two sub-groups of 7th and 21st days. The number of groups (n=6) was 8 in total. GSE of 300 mg/kg/day were provided to the subjects before they were sacrificed. LLLT of 23 J/cm2 was administered to two different points alongwith the fracture line at intervals of 48 hours for 7 days in the 7th day sub-groups and for 14 days in the 21st day sub-groups. Afterthe procedure, while biochemical values such as TAS, TOS and OSI are measured; histopathologically it was examined in termsof capillary number and width, inflammatory cell, fibroblast count, collagen fibers, osteoblast count, ossification and mature boneformations.Results: For biochemical analyses, there was statistically significant difference only in TAS values on either the 7th or 21st day forthe groups. Histological analyses showed that mandibular fracture healing were significantly better in the GSE and GSE+LLLT groupscompared to the control group. The group of only LLLT had limited recovery while the combination of GSE+LLLT was the best forossification.Conclusions: It is concluded that GSE may be one of the potential methods to accelerate fracture healings in mandibular fracturescommon in oral and maxillofacial surgery clinic and thus help patients recover in shorter time, and however LLLT can have positiveeffects on the process of ossification and recovery only when combined with the extract, which may result in better outcomes inclinical use.Öğe Kanser hücreleri üzerine Origanum Minutiflorum’un Sitotoksik Etkisinin Araştırılması(2019) Özkan, Oktay; Özbey, Caner; Aydemir, Işıl; Sarı, İsmail; Arat, Gonca Dönmez; Kırık, Fatma Esin; Savran, AhmetAmaç: Kanser, doku ve organları istila eden ve vücudun diğer kısımlarına sıçrayabilen, hücrelerin anormal gelişimidir. Meme kanseri en çok tekrarlanan tanıya sahip ve dünya genelinde kadınlarda kanserle ilişkili ölümlerde en önde gelen sebeplerdendir. Tıbbi bitkiler bazı kanser türlerinde antikarsinojenik özelliklere sahip olabilmektedir. Bu çalışmanın amacı Origanum minutiflorum O. Schwarz & P.H. Davis’in in vitro şartlarda, meme kanseri hücre dizilerinde sitotoksik etkisini araştırmaktır.Gereç ve Yöntem: Bu çalışmada, MDA-MB-231 ve MCF-7 meme kanseri hücre dizileri ve adipoz dokudan izole edilmiş mezenkimal kök hücreleri kullanıldı. Origanum minutiflorum’dan elde edilmiş esansiyel yağa maruz bırakılmış tüm hücre dizilerinde MTT testi aracılığıyla IC50 dozu hesaplandı. Her bir hücre dizisinin IC50 dozunda eNOS, p53, PCNA ve TUNEL için immunositokimya boyaması yapıldı. Sonuçlar Graphpad yazılımı kullanılarak one-way-ANOVA kullanılarak değerlendirildi.Bulgular: Meme kanseri hücrelerinin proliferasyonu oregano yağı tarafından inhibe edildi. Oregano yağı mezenkimal kök hücrelerinde herhangi bir sitotoksik etki göstermedi. eNOS ve p53 ile apoptozisin göstergesi TUNEL’de artış yoluyla oregano yağının kanser hücrelerinde sitotoksik olduğu ortaya çıktı ve PCNA boyamada proliferasyonun azaldığı tespit edildi.Sonuç: Bizim verilerimize göre oregano yağı MDA-MB-231 ve MCF-7 meme kanseri hücrelerinde sitotoksik bir etkiye sahiptir ve diğer biyolojik aktiviteleri için aktif komponentlerini tanımlayacak testlere ihtiyaç vardır.Öğe The Inhibition Effect of Curcumin on MCF-7 Breast Cancer Cells via GSK-3beta and VEGF Signals(2021) Aydemir, IşılObjective: Nowadays, there is a tendency towards supportive and alternative therapies in cancer treatment. So, theaim was to search the effect of curcumin on the cell viability and migration via GSK-3beta and VEGF in MCF-7breast cancer cells and L929 fibroblast cells.Materials and Methods: In the experiment, MCF-7 breast cancer cells and L929 fibroblast cells were treated withcurcumin at five concentrations (5, 10, 20, 40 and 80µM) for 24 and 48 hours, and MTT assay was used to detect thecytotoxicity level of curcumin. For immunocytochemical staining, both cells were exposed with 5µM, 20µM and80µM of curcumin for 48 hours. The immunocytochemistry method was performed to evaluate the expressions ofGSK-3beta and VEGF. The immunocytochemical results were evaluated using H-score. For cell migration, the scratchwound assay was done at three concentrations for 48 hours, and the percentage of wound closure was calculated. Alldata were analyzed statistically.Results: After MTT assay, it was observed that curcumin had a dose-dependent toxic effect on MCF-7 cancer cellscompared to L929 cells. The immunocytochemical distributions of GSK-3beta and VEGF were significantlydecreased with the curcumin treatment in MCF-7 cells. However, curcumin showed a marked inhibitory effect on thecell migration in comparison with the non-treated MCF-7 cells and L929 cells.Öğe The Inhibitory Effect of Capparis Ovata Polysaccharides on Cultured Pterygium Fibroblasts(2021) Küçük, Erkut; Zor, Kürşat; Aydemir, Işıl; Özkan, OktayObjectives: Pterygium recurrence after removal surgery is an important problem. Polysaccharides obtained from Capparis species have been shown to possess various biological properties, including anti-tumor activity. This study was aninvestigation of the effect of Capparis ovata polysaccharides on cultured pterygium fibroblasts and a comparison with the effects of mitomycin C (MMC).Methods: Pterygium tissue samples were obtained during excision surgery from 3 patients with primary pterygium, and fibroblasts were isolated. Pterygium fibroblast cultures and L929 cell cultures were treated with different concentrations of MMC and Capparis ovata polysaccharides. Cell proliferation and migration were evaluated.Results: An MTT assay revealed that cell viability decreased with increasing concentrations of Capparis ovata polysaccharides in both cell types. MMC also inhibited the proliferation of both cell types. A scratch-wound assay indicated thatboth Capparis ovata polysaccharides and MMC molecules reduced proliferation and migration in the pterygium fibroblasts and L929 cells.Conclusion: The in vitro Capparis ovata polysaccharide inhibition of proliferation and migration of pterygium fibroblasts was similar to that of MMC. The results of this study suggested that Capparis ovata polysaccharides may be a valuable candidate drug to treat pterygium.
