Özet
Bu çalışmada Niğde Akkaya Gölü'nden izole edilen anaerobik fotosentetik Rhodopseudomonas sp. 51ATA suşunun ışıklı-havasız ve karanlık-havaya açık koşullarda bakır metali üzerindeki korozyon aktivitesi araştırılmıştır. Bakterinin korozyon davranışı, bakteri içermeyen kontrol ortamıyla kıyaslanarak zamana bağlı olarak elektrokimyasal teknikler (polarizasyon direnci, tafel eğrileri ve A.C. impedans ölçümleri) ile ölçülmüştür. Spektrofotometrik ve koloni sayım metoduyla bakteri sayısı belirlenmiş, elektrot yüzeyindeki korozyonun derecesi fotoğraflanarak gösterilmiştir. Sonuç olarak; ışıklı-havasız koşullarda besiyer ortamında bulunan iyonların bakırla tepkimeye girerek yüzeyi kapattığı, bakteri içeren ortamda ise bakteri sayısı ile korozyon hızının paralel olarak arttığı görülmüştür. Karanlık-havaya açık koşullarda yapılan çalışmalarda ise, besiyer ve bakteri içeren ortamlarda benzer sonuçlar elde edilmiş, çalışılan her iki ortamda da zamanla korozyon akımları azalmıştır. Bu durum, kullanılan besiyer çözeltisinde bulunan iyonların metal ile etkileşerek yüzeyde birikintiler oluşturması ve bakterili ortamda bakterilerin biyofilm oluşturarak yüzeyi kapatması ile açıklanabilir.
In this study, the anaerobic photosynthetic Rhodopseudomonas sp. 51ATA strain isolated from Niğde Akkaya Lake was investigated for its corrosive effects on copper metal at light-anaerobic and dark-aerobic conditions. The corrosion behavior of bacterium was compared with the control medium (without bacterium) as a function of time by using electrochemical techniques (polarization resistance, tafel slopes and A.C. impedance measures). The number of bacterium was determined by spectrophotometric and colony unit forming methods, the degree of corrosion on the electrode surface was also photographed. As a result, the ions in the light-anaerobic medium have covered the surface by reacting with copper and the number of bacterium has increased in parallel to the corrosion rate in the medium containing bacterium. Similar results were obtained under dark-aerobic conditions for both media (with and without bacterium), corrosion currents within both media has decreased. This result can be explained by the interaction of the ions in medium with the metal to form deposits on the surface and by forming biofilm on the surface.
