Özet
Araştırmada Pseudomonas fluorescens ssp. indologenes, Pseudomonas vesicularis, Pseudomonas luteola, Pseudomonas aeruginosa türlerine ait toplam 15 adet suş kullanılmıştır. Suşlar çiğ süt örneklerinden izole edilmiş ve Analitik Profil İndeksin kullanımı (API 20NE) ile tanımlanmışlardır. Pseudomonas spp. suşlarının amfisilin (10 ?g), amikasin (30 ?g), gentamisin (10 ?g), oflaksasin (5 ?g), tetrasiklin (30 ?g), kloramfenikol (30 ?g), sefuroksin (30 ?g) antibiyotiklerine karşı duyarlılık testleri disk difüzyon yöntemine göre yapılmış, sonuçlar NCCLS kriterlerine göre değerlendirilmiştir. Pseudomonas spp. suşlarının kullanılan 7 adet antibiyotiğe karşı ortalama olarak %57'sinin duyarlı, %35'inin dirençli ve %7'sinin orta duyarlı olduğu tespit edilmiştir.Pseudomonas spp. suşlarının Skim Milk Agar (SMA) besi ortamında proteolitik aktiviteleri incelenmiş ve 14 adet suşun proteolitik aktiviteye sahip oldukları belirlenmiştir. Bu suşlarda tespit edilen proteolitik aktivite, 8,0-55,0 mm zon çapı arasında, ortalama olarak 28,1 mm hesaplanmıştır. En yüksek proteolitik aktivite zon çapı, 55,0 mm ile P. fluorescens ssp. indologenes T7'de bulunmuştur.Çalışmada, Pseudomonas spp. suşlarının Tribütirin Agar (TA) besiortamında lipolitik aktiviteleri incelenmiştir. Toplam 13 adet suşun lipolitik aktiviteye sahip oldukları belirlenmiştir. Bu suşlarda tespit edilen lipolitik aktivite 5,3-29,3 mm zon çapı arasında bulunmuş, ortalama olarak 15,4 mm hesaplanmıştır. In the study, a total of fifteen strains belonging to Pseudomonas fluorescens ssp. indologenes, Pseudomonas vesicularis, Pseudomonas luteola, Pseudomonas aeruginosa were analysed. Strains were isolated from the raw milk and identified by using Analytical Profile Indeks (API 20 NE). Antibiotic susceptibility tests of Pseudomonas spp. strains to ampicillin (10 ?g), amikacine (30 ?g), gentamicine (10 ?g), oflaxacine (5 ?g), tetracikline (30 ?g), chloramphenicol (30 ?g), cephuroxime (30 ?g) were determined by using the disk diffusion method. The results were described according to NCCLS standards. It was determined that 57% of Pseudomonas strains were susceptibility, 35% were resistant and 7% were intermediate susceptibility against fifteen antibiotics tested.Proteolytic activities of Pseudomonas isolates were tested in Skim Milk Agar (SMA) medium and fourteen strains were determined that had a proteolytic activities.
