Biber kalluslarında farklı azot konsantrasyonlarının kallus büyümesi ve kapsaisin birikimi üzerine etkisinin incelenmesi

Özet
Bu çalışmada, farklı azot konsantrasyonlarının Samandağ biberi tohumlarına ait kallus hücre kültüründe, bazı fizyolojik ve biyokimyasal parametreler üzerine etkisi araştırılmıştır. Bu amaçla standart MS besiyerinin azot miktarının % 0, % 50, % 100 ve % 150 değişen oranları kullanılarak besyerleri hazırlanmıştır. In vitro koşullarda çimlendirilmiş biber fidelerinin hipokotil eksplantlarından çimlenme ortamı ile aynı oranda hazırlanan besi yerlerine büyüme düzenleyiciler (1 mg/l 2,4 D ve 0,1 mg/l kinetin) eklenerek kallus kültürleri oluşturulmuştur. Gelişen kalluslardan örnek alınarak analiz işlemleri yapılmıştır. Farklı azot konsantrasyonlarının toplam fenolik bileşik miktarı, toplam protein miktarı değişimleri, kapsaisin miktarı ile antioksidant enzimlerden süperoksit dismutaz (SOD) ve katalaz (CAT) enzim aktiviteleri üzerine olan etkileri incelenmiştir. Elde edilen veriler doğrultusunda, artan azot konsantrasyonuyla paralel şekilde kapsaisin miktarını arttığı tespit edilmiştir. Kontrol ile karşılaştırıldığında azot kaynağının azaltıldığı ortamda toplam fenolik madde miktarındaki artış en yüksek bulunmuştur. Azot oranlarının değiştirildiği ortamların tümünde standart MS besiyerinin kullanıldığı gruba oranla toplam protein miktarında azalmalar, SOD ve CAT enzim aktivitelerinde artışlar meydana gelmiştir.
This study investigated the effects of different nitrogen concentrations on some physiological and biochemical parameters in the callus cell culture of Samandağ pepper seeds. To achieve this, the media were prepared using varying ratios of 0%, 50%, 100%, and 150% nitrogen of standard MS medium. Callus cultures were formed by adding growth regulators (1 mg/l 2,4 D and 0.1 mg/l kinetin) to the media prepared at the same rate as the germination medium from hypocotyl explants of germinated pepper seedlings in vitro. Samples from the growing calli were used for the analysis methods. The effects of different nitrogen concentrations on the total amount of phenolic compounds, the changes in the total protein content, the amount of capsaicin, and the activities of antioxidant enzymes superoxide dismutase (SOD) and catalase (CAT) were investigated. According to the data collected, it was found that the capsaicin content increased in step with the rising nitrogen concentration. The increase in total phenolic substance was found to be highest in the medium where the nitrogen source was reduced compared to the control. In all media in which nitrogen ratios were changed, there were decreases in total protein amount and increases in SOD and CAT enzyme activities compared to the group in which standard MS medium was used.
