Özet
Bu çalısmada bazı dogal boyaların, solüsyondaki ve tekstil materyali üzerindeki antimikrobiyal aktivitesi arastırılmıstır. Kök boya (Rubia tinctorum L.), sogan (Allium cepa L.), nar (Punica granatum L.) ve nane (Mentha piperita L.) bitkilerinden elde edilen 4 adet dogal boya Staphylococcus aureus ATCC 25923, Shigella sonnei RSKK 877, Escherichia coli ATCC 35218, Bacillus megaterium RSKK 5117, Bacillus subtilis RSKK 244, Bacillus cereus RSKK 863, Pseudomonas aeruginosa ATCC 29212, S.epidermidis, Salmonella 21.3, Pseudomonas aeruginosa ATCC 27853 bakterilerine karsı disk difüzyon yöntemi ile test edilmistir. Çalısmada kullanılan boyalardan (100 ?l'de) nar kabugunun 8 adet test bakterisi üzerinde10,5?17,9 mm zon çapı aralıgı ile kökboyanın 3 adet test bakterisi üzerinde 8,5?10,2 mm zon çapı aralıgı ile nanenin 6 adet test bakterisi üzerinde 10,2?13,5 mm zon çapı aralıgı ile ve sogan kabugunun 3 adet test bakterisi üzerinde 9,9- 13,5 mm zon çapı aralıgı ile inhibisyon aktivitesi gösterdigi belirlenmistir. Test bakterileri üzerinde en yüksek inhibisyon aktivitesine sahip olan boyanın nar kabugu oldugu tespit edilmistir. Disk difüzyon çalısması sonucunda test bakterilerine karsı antimikrobiyal aktivite gösteren boyalar ve etki ettikleri bakteriler seçilmis ve bu boyalarla boyanmıs yün ip numunelerinin antimikrobiyal aktivitesi test edilmistir. Buna göre nar kabuguyla boyanmıs yün ipte % bakteri indirgenmesinin % 4? 80; sogan kabuguyla boyanmıs yün ipte % 53?86; kök boyayla boyanmıs yün ipte %32?52; nane bitkisiyle boyanmıs yün ipte % 28?91 arasında degisiklik gösterdigi belirlenmistir. Çalısmada nar ve nane bitkileri dogal boyalarının test edilen bakterilere karsı maksimum inhibisyon zonu ve en iyi mikrobiyal indirgenmeyi gösterdigi tespit edilmistir. In this study, antimicrobial activity of some natural dyes in solution and antimicrobial activity on textile material was investigated. Four natural dyes madder (Rubia tinctorum L.), onion (Allium cepa L.) , pomegranate (Punica granatum L.), mint (Mentha piperita L.) were tested against Staphylococcus aureus ATCC 25923, Shigella sonnei RSKK 877, Escherichia coli ATCC 35218, Bacillus megaterium RSKK 5117, Bacillus subtilis RSKK 244, Bacillus cereus RSKK 863, Pseudomonas aeruginosa ATCC 29212, S.epidermidis, Salmonella 21.3, Pseudomonas aeruginosa ATCC 27853 by using disc diffusion method. Inhibition activitiy of pomegranate naturel dyes against 8 test bacteria were determined between 10,5?17,9 mm, madder naturel dyes against 3 test bacteria were determined between 8,5?10,2 mm, mint naturel dyes against 6 test bacteria were determined between 10,2?13,5 mm and onion naturel dyes against 3 test bacteria were determined between 9,9- 13,5 mm. It was shown that pomegranate naturel dyes had the highest inhibition activity against test bacteria. In the next set of the experiments the antimicrobial activity of dyed wool specimens were tested. A reduction of 4-80 % in bacterial growth were determined on wool sample dyed with pomegranate; a reduction of 53-86 % on wool sample dyed with onion; a reduction of 32?52 % on wool sample dyed with madder; a reduction of 28?91 % on wool sample dyed with mint. Pomegranate natural dye and mint natural dye were shown maximum inhibition zone in solution and also the best microbial reduction against all the microbes tested.
