Özet
Bu çalışma su, zeytinyağı ve alkol (metanol, etanol) kullanılarak ekstrakte edilen propolisin antioksidan aktivitesi ve toplam fenolik madde değeri belirlenerek en uygun çözücünün belirlenmesi amacıyla yürütülmüştür. Propolis örnekleri Kovanlardan toplanan propolis örnekleri farklı konsantrasyonlarda (%90, %70 ve %50) metanol ve etanol ile saf su ve zeytinyağı kullanılarak ekstrakte edilmiştir. En yüksek ortalama antioksidan aktivite değeri %90 konsantrasyonunda 1/20 lik metanol grubunda gözlemlenmiş, en düşük antioksidan aktivite değeri ise 1/5 oranında su ile ekstrakte edilen grupta bulunmuştur. En yüksek ortalama toplam fenolik madde değeri %90 konsantrasyonunda 1/20 metanol ile ekstrakte edilmiş grupta bulunurken, en düşük ortalama toplam fenolik 1/5 oranındaki su grubunda gözlemlenmiştir. En iyi propolis çözücüsünün %90 lık metanol olduğu gözlenmiştir. Konsantrasyon ve metanol oranı artıkça propolisteki çözünme oranının ve buna bağlı olarakta antioksidan aktivitenin ve toplam fenolik madde değerinin arttığı görülmüştür. Ham propolisin çözülmesinde su ve yağ yerine alkol kullanılması gerektiği, mümkünse yüksek konsantrasyonda alkolle çözmenin yapıldıktan sonra alkolün evapore edilerek tekrar geri alınması ile iyi bir propolis ektraksiyon işleminin yapılabileceği önerilebilir.
This study was carried out to determine the most suitable solvent by determining the antioxidant activity and total phenolic value of propolis extracted using water, olive oil and alcohol (methanol, ethanol). Propolis samples were extracted with different concentrations (90%, 70% and 50%) of methanol, ethanol, water and olive oil (1/5, 1/10, 1/20 g / mL). When the results obtained were examined, the highest mean antioxidant activity value was observed as in 90% concentration methanol group and the lowest antioxidant activity value was observed as in the group extracted with water in 1/5 ratio. The highest mean total phenolic content was found to be in the group extracted with 1/20 methanol at a concentration of 90%, while the lowest mean total phenolic matter value was observed as in the water group of 1/5 ratio. These results showed that methanol was the best propolis solvent among the solvents used in this study and that the best solubility in the methanol group was in the 90% group. It has seen that as concentration and solvent ratio increased, dissolution rate, antioxidant activity and total phenolic value of propolis increased. It can be suggested that alcohol should be used instead of water and oil to dissolve the crude propolis, and if possible, a good propolis extraction can be performed by evaporing the alcohol after dissolving with high concentration of alcohol.
