Phenotypic and genotypic characterization of pepper genotypes for tomato spotted wilt virus (TSWV) disease reaction and resistance

Domates lekeli solgunluk virüsünün, sera koşullarında bazı yerel ve ticari biber genotiplerinin verim ve kalite parametrelerine etkilerinin araştırılması için mekanik inokulasyonlar yapılmış ve inokulasyonlardan sonra virus enfeksiyonu RT-PCR ve DAS-ELISA ile test edilmiştir. DAS-ELISA sonuçlarına göre inokule edilen bitkilerin %95' inin TSWV pozitif olduğu tespit edilirken, RT-PCR sonuçlarına göre ise istenilen uzunlukta (276 bç) bantlar elde edilmiştir. TSWV enfeksiyonu, virüsle inokule edilen bitkilerde verim (%68,62-100), meyve sayısı (%45,97-100), meyve ağırlığı (%66,38-98,77), meyve eni (%63,77-96,52), meyve boyu (%31,97-93,514), meyve eti kalınlığı (%28,64-82,41), meyve sertliği (%5,82-94,43), meyve rengi (%1,62-7,79) gibi kalite parametrelerinde önemli (p?0.05) düşüşlere neden olmuştur. Sonuçlara göre, TSWV enfeksiyonuna karşı en iyi performansı "Yalova Çarliston 341" biber çeşidi gösterirken, en çok etkilenen çeşidin ise "Bora 77" olduğu görülmüştür. Ayrıca, Tsw dayanıklılık geninin tespiti için SCAC568 markörü ve TaqI kesimleme enzimi kullanılarak PCR testleri yapılmış, ancak C. chinense yabani biber genotipi ile dayanıklı olduğu bilinen "153-227" dışındaki çeşitlerin Tsw genine sahip olmadığı tespit edilmiştir.
To determine the effects of Tomato spotted wilt virus (TSWV) on the yield and quality parameters of some local and commercial pepper genotypes under greenhouse conditions, mechanical inoculations were performed and virus infections were tested by RT-PCR and DAS-ELISA after inoculations. Based on the DAS-ELISA results, 95% of the inoculated plants were positive for TSWV, in accordance with the expected amplicon size (276 bp) obtained by RT-PCR results. Infection of TSWV caused significant (p?0.05) reduction in fruit number (45,97-96,37%), fruit weight (66,38-98,77%), fruit width (63,77-96,52%), fruit lenght (31,97-93,514%), flesh thickness (28,64-82,41%), fruit firmness (5,82-94,43%), fruit color (1,62-7,79%), and yield (68,62-98,85%) in inoculated plants. As a conclusion, the best performance against TSWV was observed on cv. Yalova Çarliston 341, while the least one observed on cv. Bora 77. Moreover, incidence of Tsw resistance gene was investigated by PCR using SCAC568 marker and TaqI restriction enzyme; however Tsw gene could not be detected in any of the tested cultivars except wild type C. chinense and resistant genotype cv.153-227.
