Özet
Bu çalışmanın amacı, Lactobacillus plantarum HP1 bakterisinin in vitro koşullarda probiyotik özellikleri belirleyerek probiyotik bakteri olarak kullanılabilme potansiyelini ortaya koymaktır. Bu amaçla Lb. plantarum HP1'in düşük asitliğe, safra tuzuna, pepsin, pankreatin, yapay mide sıvısı, fenol ve antibiyotiklere karşı direnci belirlenmiştir. Ayrıca; hemolitik aktivite, antimikrobiyal aktivite, otoagregasyon, koagregasyon ve hidrofobisite özellikleri de tespit edilmiştir. Araştırma sonucunda, Lb. plantarum HP1'in hemoliz negatif, düşük asitliğe, safra tuzuna, pepsin, pankreatin, yapay mide sıvısı ve fenole dayanıklı olduğu ve test edilen 11 antibiyotikten sadece birine duyarlı olduğu, otoaggregasyon, koaggregasyon ve hidrofobisite özelliklerini iyi olduğu belirlenmiştir. Ayrıca patojen bakterilerden Enterococcus faecium, E. feacalis, Staphylococcus aureus, Bacillus cereus, Listeria monocytogenes ve L. ivanovii'ya karşı inhibitör aktiviteye sahip olduğu gözlenmiştir. Sonuç olarak, Lb. plantarum HP1 bakterisinin gıda endüstrisinde probiyotik olarak kullanma potansiyeline sahip olduğu söylenebilir.
The aim of this study was to determine the probiotic properties of Lactobacillus plantarum HP1 bacterium in vitro condition and to reveal its potential to be used as a probiotic bacterium. For this purpose, Lb. plantarum HP1's resistance to low acidity, bile salt, pepsin, pancreatin, artificial gastric juice, phenol and antibiotics was determined. In addition, hemolytic activity, antimicrobial activity, autoaggregation, coaggregation and hydrophobicity properties were also determined. As a result of the research, Lb. plantarum HP1 was hemolysis negative, resistant to low acidity, bile salt, pepsin, pancreatin, artificial gastric juice and phenol, and was sensitive to only one of the 11 tested antibiotics, with good autoaggregation, coaggregation and hydrophobicity properties. It has also been observed to have inhibitory activity against pathogenic bacteria Enterococcus faecium, E. feacalis, Staphylococcus aureus, Bacillus cereus, Listeria monocytogenes and Listeria ivanovii. As a result, Lb. plantarum HP1 bacterium has the potential to be used as a probiotic in the food industry.
